Experience of Magento is a must.

Designers need to be conversant with Adobe Suite CS 3/4, Photoshop, Illustrator, Dreamweaver, and ideally, flash. Hand coding (x)HTML and CSS. Experience of being able to take initial designs and convert them into Magento templates and themes is required.

Developers need CSS, (x)HTML, JQuery, Ajax, PHP, MySQL and SQL skills. Experience of customisations and creating extensions would give you the edge.
